Description
Reslizumab (Sch 55700) is a humanized immunoglobulin G (IgG)4 κ monoclonal antibody that binds to human interleukin 5, thereby decreasing eosinophil production and survival.Reslizumab improves lung function, induces a decrease in eosinophil production and maturation, and may be used in the study of asthma.
Targets & IC50
IL-5 (human):109 pM (KD), IL-5 (human):20 pM (Kd), IL-5:91.1 pM, Ba/F3 cells:0.5 nM
In vitro
Reslizumab binds with high affinity to human IL-5 in Biacore surface plasmon resonance, KD = 109 pM and in kinetic exclusion assays, KD = 4.3 pM. [1]
Reslizumab displays high affinity binding to hIL-5 (Kd = 20 pmol/l), inhibits the binding of hIL-5 to Ba/F3 cells (IC50 = 0.5 nmol/l) and blocks IL-5 mediated proliferation of human erythroleukemic TF-1 cells. [2]
In vivo
Intraperitoneal or intramuscular injection of 0-10 mg/kg Reslizumab inhibited pulmonary eosinophilia in allergic mice in a dose-dependent manner. [2]
Intravenous injection of 30 mg/kg Reslizumab inhibited allergic bronchoconstriction but not histamine-induced bronchoconstriction in allergic guinea pigs. Intravenous injection of 0.1-1 mg/kg Reslizumab blocked pulmonary eosinophilia and neutrophilia induced by tracheal injection of hIL-5 in guinea pigs. [2]
Intravenous 0.3 mg/kg Reslizumab blocked antigen-challenge-induced pulmonary eosinophilia for up to 6 months in allergic rhesus monkeys. [2]
