Description UBE2T/FANCL-IN-1 is a highly effective inhibitor of UBE2T/FANCL-mediated FANCD2 monoubiquitylation. It greatly enhances the sensitivity of cells to Carboplatin, a DNA cross-linking agent.
In vitro UBE2T/FANCL-IN-1 (CU2) effectively prevents GST-FANCLRING ubiquitylation at a concentration of 100 μM. At a higher dosage of 500 μM, it significantly reduces the ubiquitylation of FANCD2 mediated by HU and Cisplatin (10 μM) treatments over timespans of 6, 12, and 24 hours. Furthermore, using 250 μM of UBE2T/FANCL-IN-1 in combination with 15 μM carboplatin markedly inhibits cell proliferation, resulting in less than 50% confluence after 8 days[1].
