SHMT2-IN-1 is a potent inhibitor of serine hydroxymethyltransferase 2 (SHMT2) with an IC50 of 1.21 μM and a KD of 10.6 μM. It binds to the folate binding site of SHMT2 and also inhibits SHMT1 with an IC50 of 1.83 μM. MT-44 can suppress the proliferation, migration, and invasion of cancer cells, inducing apoptosis and reactive oxygen species (ROS) production, leading to G2/M phase arrest. SHMT2-IN-1 is applicable in cancer research, including studies on esophageal cancer.

| In vitro | SHMT2-IN-1 exhibits potent antiproliferative effects against esophageal cancer cells KYSE-450 and KYSE-70, with IC50 values of 1.62 μM and 1.18 μM, respectively, at concentrations ranging from 0.1 to 10 μM over 24 hours. Additionally, SHMT2-IN-1, at concentrations between 2.5 and 10 μM over 24 hours, inhibits the migration and invasion of these cells in a dose-dependent manner. Moreover, SHMT2-IN-1, used at 2.5 to 7.5 μM for 24 to 72 hours, induces apoptosis, generates reactive oxygen species, and causes G2/M phase arrest in KYSE-450 and KYSE-70 cells. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
