Description
S1R agonist 3 is a potent and selective sigma-1 receptor (S1R) agonist with a Ki of 1.5 nM. Additionally, S1R agonist 3 can reverse learning and memory impairment induced by Aβ25-35. S1R ligand 1 is applicable for Alzheimer's disease research.

In vitro
S1R agonist 3 (Compound 1) binds effectively to the S1R on transfected HEK293 cell membranes with a K i value of 1.5 ± 0.5 nM at concentrations ranging from 0.01-10000 nM over 120 minutes. Additionally, S1R agonist 3 acts as a potent S1R agonist in overexpressed CHO cells, inducing S1R-BiP dissociation with an EC 50 of 160 ± 8 nM at concentrations of 0.01-1 μM over 30 minutes.
In vivo
S1R agonist 3 (Compound 1), at a concentration of 3 μM over 24 hours, effectively suppresses hyperactivity in wfs1abKO zebrafish larvae without affecting the normal movement of wild-type larvae. Additionally, S1R agonist 3 administered intraperitoneally at 0.03 mg/kg as a single dose, 20 minutes after Aβ 25-35 injection, strongly reverses learning and memory deficits induced by Aβ 25-35 in mice.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
