Rev1-CT/RIR PPI-IN-2 (compound 10) is a translesion synthesis (TLS) inhibitor targeting the interaction of the Rev1 protein, with an IC50 of 13.7 μM and Kd of 40.8 μM, exhibiting anticancer properties. It enhances cisplatin-mediated cytotoxicity by disrupting the interaction between the C-terminal domain of TLS DNA polymerase Rev1 (Rev1-CT) and the Rev1 interaction region (RIR) of other TLS DNA polymerases.

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
