PMEG is an inhibitor targeting the nuclear DNA polymerases α, δ, and ε (DNA polymerases α/δ/ε), leading to DNA chain termination and suppression of DNA synthesis, which induces cytotoxicity in proliferating cells. This acyclic nucleoside phosphonate converts intracellularly to its active phosphorylated metabolite, PMEG diphosphate. In rodent models, PMEG demonstrates activity against leukemia and melanoma. Due to its poor cellular permeability, its prodrug form is known as Rabacfosadine (GS-9219). PMEG exhibits antiviral activity against several DNA virus infections, including murine cytomegalovirus (MCMV) and human cytomegalovirus (HCMV). It is also investigated for non-Hodgkin's lymphoma research.

Description

In vivo
Rabacfosadine (GS-9219) administered intravenously at doses of 0.20-0.82 mg/kg over 30 minutes can significantly accumulate PMEG in lymphoid cells, leading to a remission rate of 79% in domestic dogs with spontaneous non-Hodgkin's lymphoma.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
