| In vitro | PI3K/PIKK-IN-1 (Compound 1) exhibits potent inhibition of recombinant human FRAP1 (mTOR) kinase activity in vitro with an IC50 of 3.6 nM when used in concentrations ranging from 0.000051 μM to 3 μM for 1 hour. It also strongly inhibits recombinant human PIK3CA/PIK3R1 (p110α/p85α) kinase activity with an IC50 of 0.5 nM at concentrations of 0.046-100 nM for 1 hour. Moreover, PI3K/PIKK-IN-1 significantly suppresses the proliferation of MDA-MB-453 and MDA-MB-468 breast cancer cells under in vitro conditions at concentrations of 0.015-250 nM over a period of 6 days. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
