In vitro
PD166285 demonstrates significant antiproliferative effects in CCNE1-amplified OVCAR3 cells (IC 50 = 0.14 μM) and HCC1569 cells (IC 50 = 0.21 μM) when applied at concentrations ranging from 0.062 to 1 μM over 5 days. It also inhibits colony formation in a dose-dependent manner in the same cells over 14 days, with an EC 50 of 0.06 μM for OVCAR3 cells and 0.16 μM for HCC1569 cells. Additionally, PD166285 induces apoptosis and causes arrest of CCNE1-amplified HCC1569 cells at the G1/S phase of the cell cycle when used at concentrations of 0.125 to 1 μM over 5 days.
