| In vitro | NSC 727357, at concentrations below 10 μM, induces Top1 cleavage complex formation and at higher levels (≥10 μM), it inhibits Top1 catalytic activity, while also causing a minor induction of Top2-DNA cleavage complexes. This compound intercalates into DNA and induces supercoiling. At concentrations of 0.1-5 μM over 18 hours, NSC 727357 causes G1 phase arrest in HT29 cells. It exhibits potent antiproliferative activity against the NCI-60 cell line panel, with a GI50 value of 0.067 μM. |

1) Add 100 μL of the DMSO stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
