Description
NEP162 is a BRD4 PROTAC degrader with DC50 values of 1.2 μM in SW480 cells and 1.6 μM in U2OS cells. NEP162 exhibits antiproliferative activity, effectively inhibiting tumor growth and inducing apoptosis. It is applicable in research on cancers such as osteosarcoma, colorectal cancer, and non-small cell lung cancer.
In vitro
NEP162 (2 μM, 12-18 h) degrades BRD4 through GID4 and UPS in SW480, SW480 cells with siRNA knockdown, and GID WT cells. NEP162 (2 μM, 0-72 h) inhibits proliferation, induces apoptosis, and affects colony formation in U2OS cells.
In vivo
NEP162 (5-15 mg/kg, i.p., once every 2 days, for 14 days) inhibited tumor growth in a mouse xenograft model constructed using U2OS cells.
