| Description | NEK7degrader-3 is an orally active NEK7 molecular glue degrader capable of crossing the blood-brain barrier, with a DC50 of 31.1 nM. It facilitates the interaction between NEK7 and the E3 ligase cereblon, promoting NEK7 proteasomal degradation and reducing inflammation mediated by the NLRP3 inflammasome. NEK7degrader-3 inhibits caspase-1 activity, releases cytokines IL-1β, IL-1α, and IL-18, and decreases pyroptosis-related plasma membrane permeability. It shows anti-inflammatory effects in an LPS-induced neuroinflammation mouse model and is useful for studying neuroinflammation. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
