In vitro
MPXV p37 protein-IN-1 (compound C12) exhibits potent anti-MPXV virus activity in Vero cells at a concentration of 1-10 μM over 48 hours. It forms essential hydrogen bonds with residues TYR258 and SER292, which may enhance its binding affinity and antiviral efficacy.
In vivo
Compound C12, also known as MPXV p37 protein-IN-1, is well-tolerated in CD-1 mice at a single oral dose of 2000 mg/kg. Additionally, a daily oral dose of 100 mg/kg administered over 14 consecutive days is also well-tolerated in the same mouse model.
