KWCN-41 is a potent and selective RIPK1 kinase inhibitor (IC50=88 nM) that specifically inhibits necrosis without affecting apoptosis and exhibits anti-inflammatory effects.

In vitro
KWCN-41 (10/50/50 nM; 1 h) dose-dependently inhibited necrosis in L929 cells [1].
KWCN-41 (10/50/250 nM; 0.5/1.5/2.5 h) inhibited phosphorylation of RIPK1/3 and MLKL in L929 cells [1].
KWCN-41 (0-10 μM) dose-dependently inhibited TZ-induced necrosis in L929 cells and suppressed TSZ-induced necrosis in HT-29 and U937 cells [1].
In vivo
KWCN-41 (10/25/40 mg/kg; intraperitoneal injection; 72 hours) dose-dependently inhibited TNF-α-induced mouse mortality and improved survival rates [1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
