| Description | IPA is a specific Sec18/NSF inhibitor. It binds competitively to phosphatidic acid, inducing conformational rigidity in Sec18/NSF, which disrupts its essential flexibility and blocks SNARE activation and vacuole membrane fusion. IPA can be utilized to study the activity of Sec18 and the dynamics of SNARE activation. |
| In vitro | IPA binds to purified monomeric Saccharomyces cerevisiae Sec18, as well as the isolated Sec18 D1 and D2 domains, including the D1-D2 recombinant, with the highest binding affinity observed in the D1 domain [IC 50 = 762 nM (Sec18), 220 nM (D1), 641 nM (D2)]. IPA effectively and specifically competes with phosphatidic acid for binding to monomeric Saccharomyces cerevisiae Sec18 and its isolated D1 and D2 domains, without significantly inhibiting the interaction between phosphatidic acid and other phosphatidic acid-binding proteins or the association of Sec18 with PI liposomes. At a concentration of 100 μM, IPA strongly inhibits the binding of monomeric Saccharomyces cerevisiae Sec18 to PA liposomes with varying membrane curvature, showing a slightly stronger inhibitory effect on larger diameter liposomes. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
