Description
Influenza A virus-IN-18 (Compound C2e) is an orally active inhibitor targeting the Influenza A virus, displaying an IC50 of 2.5 μM against PR8/H1N1 and 6.42 μM against H3N2. This compound demonstrates high-affinity binding directly to the H1N1/PR8 HA protein with a KD of 0.25 μM. Influenza A virus-IN-18 is applicable for research on Influenza A virus infections.
In vitro
Influenza A virus-IN-18 binds directly to the H1N1/PR8 HA protein with high affinity, exhibiting a K D of 0.25 μM at concentrations ranging from 1.25 to 20 μM. Over a 48-hour period, Influenza A virus-IN-18 effectively inhibits replication of the influenza A virus PR8/H1N1 in MDCK cells, with an IC 50 of 2.5 μM. Additionally, for H3N2 virus replication in MDCK cells, Influenza A virus-IN-18 shows an IC 50 of 6.42 μM.
In vivo
Influenza A virus-IN-18, administered orally at a dose of 2.5-5 mg/kg/day once daily, resulted in over 50% survival rate in H1N1-infected BALB/c mice, and showed superior weight recovery compared to control treatment. This compound was well tolerated at a single dose of 1000 mg/kg in ICR mice, with no observed toxicity or organ damage.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
