Description
IGN523 is an anti-CD98 antibody (hCD98, KD = 0.55 nM) that induces antibody-dependent cell-mediated cytotoxicity (ADCC). IGN523 permeabilizes lysosomal membranes and inhibits essential amino acid transport function, leading to caspase-3 and caspase-7 mediated apoptosis of tumor cells. IGN523 inhibits tumor growth in various tumor xenograft models and is used in cancer biology research studies focusing on CD98-mediated amino acid transport, lysosomal membrane stability, and antibody effector function mechanisms in tumor xenograft systems.
In vitro
Methods:The ADCC activity of IGN523 against Ramos, KG-1 and OCI-AML-3 cells was detected. Cells were treated with 2 µg/mL IGN523 for 0.5–48 hours to assess changes in cell apoptosis, lysosomal membrane permeability and amino acid transport function.
Results:
1.IGN523 induced remarkable ADCC activity in the above cell lines, with EC₅₀ values of 18.6 ng/mL, 73.8 ng/mL and 83.3 ng/mL respectively, and its ADCC activity remained stable.
2.Treatment with 2 µg/mL IGN523 for different durations triggered caspase-dependent apoptosis, increased lysosomal membrane permeability and inhibited amino acid transport [1].
In vivo
Methods:Xenograft tumor-bearing mice were intraperitoneally injected with 15 mg/kg IGN523 once a week for different treatment cycles, and tumor growth was monitored.
Results:
1.Administration once a week for 2 to 3 cycles effectively inhibited the growth of Ramos and Dau xenografts.
2.Dosing once a week for 2 to 4 cycles also exerted prominent anti-tumor effects on AML and primary NSCLC xenografts [1].
