IAV-IN-2 (Compound MC-22) inhibits influenza A virus (IAV) by preventing its entry into host cells via clathrin-mediated endocytosis (CME).

| In vitro | IAV-IN-2, at concentrations ranging from 0.01 to 10 µM, effectively inhibits IAV strains A/X-31 (H3N2) with an IC₅₀ of 4.08 µM, A/WSN/33 (H1N1), A/Udorn/72 (H3N2), and A/NYMC (H3N2), achieving inhibition rates between 97% and 100%. Meanwhile, at concentrations of 1 to 1000 μM, IAV-IN-2 exhibits cytotoxicity in A549 cells with a CC₅₀ of 33.9 μM. Additionally, at 10 μM, IAV-IN-2 interferes with the maturation of endosomes, subsequently hindering the progressive uncoating and genomic release of viral cores during endosomal transport. |
