Description
cGAS-IN-7 is an inhibitor of the cyclic GMP-AMP synthase (cGAS) with an IC50 value of 0.66 μM for human cGAS. It moderately reduces cGAMP levels in THP1 cells and is applicable in Alzheimer's disease research.
In vitro
cGAS-IN-7 (compound 6) exhibits varying degrees of cGAS inhibition at concentrations of 10 μM and 20 μM over a period of 3 hours in cell-free assays. At concentrations of 10 μM, 30 μM, and 100 μM for 16 hours, cGAS-IN-7 significantly inhibits cGAS activity and downstream signaling in THP1 Dual cells. It reduces IFN levels but does not affect NF-κB levels at a concentration of 100 μM.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
