| In vitro | Methods: Human colorectal cancer Caco-2 cells were seeded at 2 × 10³ cells per well and cultured for 24 hours. Purified Methyl alpha-eleostearate was added and the cells were treated for 24 hours; cell viability was then assessed using the WST-1 assay. Results: Methyl alpha-eleostearate inhibited Caco-2 cell proliferation in a dose-dependent manner; at 12.5 μM, cell survival was only 19%. [1] Methods: Following treatment with Methyl alpha-eleostearate, the levels of DNA fragmentation—a hallmark of apoptosis—were detected in Caco-2 cells using an ELISA assay. Results: Methyl alpha-eleostearate significantly induced apoptosis in Caco-2 cells, resulting in characteristic DNA fragmentation. [1] Methods: DLD-1 cells were treated with 0–20 μM Methyl alpha-eleostearate or 20 μM linoleic acid/CLA for 12 hours. Phospholipid hydroperoxides (PCOOH, PEOOH) were measured by CL-HPLC, and lipid peroxidation products were determined using the TBARS method. Results: Methyl alpha-eleostearate caused a dose-dependent increase in PCOOH, PEOOH, and TBARS levels.[2] |
| In vivo | Methods: Nude mice were subcutaneously inoculated with DLD-1 cells and randomly divided into a control group and a tung oil fatty acid group (rich in Methyl alpha-eleostearate). The experimental oil (50 mg) was administered orally every 2 days for 32 days, and tumor weight, body weight, and liver weight were recorded. Results: Tumor weight in the tung oil (Methyl alpha-eleostearate) group decreased by approximately 70%; there were no significant differences in body weight or liver weight among the mice, indicating good in vivo safety. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
