1,4-Naphthoquinone-NH-C2-NH-Trolox is a compound that combines Trolox and vitamin K, known for its neuroprotective properties and ability to penetrate the blood-brain barrier. It effectively inhibits processes such as oxytosis (oxidative cell death), ferroptosis, ATP depletion, and reactive oxygen species (ROS) production. Additionally, it selectively suppresses the expression of M1 markers and can be utilized in research on neurological disorders like Alzheimer's disease (AD).

| In vitro | 1,4-Naphthoquinone-NH-C2-NH-Trolox (Compound 16e) exhibits potent and comparable radical scavenging activity, with an EC50 of 15.4 μM. It inhibits oxidative cell death, ferroptosis, and ATP depletion in HT22 hippocampal neuronal cells at concentrations of 0.38-25 μM over 24 hours. Additionally, 1,4-Naphthoquinone-NH-C2-NH-Trolox reduces reactive oxygen species formation in HT22 cells at 0.4 μM for 8 hours. At concentrations of 0.5-2.5 μM for 24 hours, it selectively inhibits M1 markers in HT22 cells. |
| In vivo | Compound 16e (1,4-Naphthoquinone-NH-C2-NH-Trolox) administered via intraperitoneal injection at a dosage of 0.1 mg/kg once daily for seven consecutive days effectively improves cognitive deficits in Alzheimer's disease mice induced by Aβ 25-35. |
