| In vitro | Method: Embryonic neural stem cells were exposed to Yhhu-3792 and evaluated by BrdU incorporation, neurosphere growth and Notch-pathway analyses. DAPT was used to examine dependence on Notch signaling. Rresult: Yhhu-3792 increased neural-stem-cell proliferation and neurosphere expansion and increased Hes3 and Hes5 expression. DAPT blocked the proliferative effect, supporting Notch-pathway dependence.[1] |
| In vivo | Method: Eight-week-old male C57BL/6 mice received chronic Yhhu-3792 administration. Neural-stem-cell proliferation, hippocampal neurogenesis, Morris water-maze performance and fear-conditioning behavior were evaluated. Rresult: Yhhu-3792 expanded the neural stem-cell pool, increased adult hippocampal neurogenesis and improved spatial and episodic memory performance.[1] |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
