Description
TRPV4/TRPA1-IN-1 is an inhibitor of TRPV4/TRPA1 that blocks calcium influx mediated by these channels. It alleviates pain behaviors in a mouse model of trigeminal stimulation pain and reduces inflammation and pain-related behaviors in a mouse model of acute pancreatitis. TRPV4/TRPA1-IN-1 is useful for the study of acute pancreatitis.

In vitro
TRPV4-IN-4 (0-10 μM) inhibits TRPV4-mediated Ca 2+ influx in TRPV4-overexpressing N2a cells, primary pig articular chondrocytes, and primary rat astrocytes, with an IC 50 of 0.45 μM in N2a cells. TRPV4-IN-4 (5 μM; 5 minutes) effectively suppresses TRPV4-mediated transmembrane currents in N2a cells expressing TRPV4-GFP+. It also inhibits TRPA1 activity in TRPA1-overexpressing N2a cells, with an IC 50 of 0.43 μM.
In vivo
TRPV4-IN-4 (10 mg/kg; i.p.; single dose) alleviates trigeminal neurogenic pain mediated by TRPV4 and TRPA1 in mouse pain models. Additionally, TRPV4-IN-4 (10 mg/kg; intraperitoneal injection; single dose) reduces inflammation and pain associated with Caerulein-induced acute pancreatitis models, effectively eliminating pancreatic edema and pain behaviors.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
