Description
Toralizumab (IDEC-131) is a humanized monoclonal antibody directed against CD40L (CD154). Toralizumab (IDEC-131) targets CD40L on T cells and inhibits CD40 signaling. Toralizumab (IDEC-131) is utilized in immunology research focused on T cell co-stimulatory pathway regulation, CD40–CD40L axis modulation, and systemic lupus erythematosus (SLE)-associated immune signaling studies.
In vitro
Methods:In vitro thrombosis assays, mixed lymphocyte reaction (MLR) and cell co-culture experiments were performed to investigate the prothrombotic effect, immunomodulatory activity and adhesion molecule regulation of toralizumab.
Results:
1.Toralizumab exhibited in vitro prothrombotic activity: it activated platelets via the FcγRIIa pathway, which was initiated by cross-linking of soluble CD154 with anti-CD154 antibody to form large immune complexes [4].
2.Single treatment with toralizumab at 0–200 μg/mL only slightly suppressed lymphocyte proliferation in the MLR system without a dose-dependent effect.
3.Toralizumab blocked the upregulation of CD62E induced by co-culture of HEC and D1.1 cells [5].
In vivo
Methods:Naive experimental monkeys received intravenous Toralizumab at 15–20 mg/kg per dose. Doses were administered on Days 0, 1, 3 and 7, followed by once-weekly dosing for 8 consecutive weeks, and animal survival was monitored.
Results:This dosing regimen moderately prolonged the survival of treated monkeys, with longer survival duration compared with untreated control animals [5].
