TK-684 is a potent and selective allosteric inhibitor of SHP2, with IC50 values of 2.1 nM for SHP2WT and greater than 1000 nM for SHP2PTP. It inhibits cell proliferation and induces apoptosis (apoptosis). Additionally, TK-684 reduces the protein expression of p-AKT and p-ERK.

TK-684 inhibits the proliferation of KYSE-150 cells when used at concentrations of 1.25-100 μM for 72 hours. It induces apoptosis at concentrations of 0-40 μM over 48 hours. Additionally, TK-684 decreases the protein expression of p-AKT and p-ERK in a dose-dependent manner at concentrations of 0-20 μM for 2 hours.
