Description
Tetrahydrofolic acid (Standard) is a reference standard for research and analysis in studies involving Tetrahydrofolic acid. Tetrahydrofolic acid (5,6,7,8-tetrahydrofolic acid) is a vitamin B9 derivative that functions as a one-carbon donor in organisms and is involved in growth and metabolism. Tetrahydrofolic acid (Standard) is utilized in biochemical research focused on folate-mediated one-carbon transfer pathways, nucleotide biosynthesis mechanisms, and metabolic flux analysis in cellular proliferation systems.
In vitro
Methods:Adh5-knockout DT40 cells were treated with gradient concentrations of tetrahydrofolic acid for 3 days and 16 hours respectively. Cytotoxicity, DNA and chromosomal damage were detected, and the aggregation of damage-related proteins was observed to explore the regulatory mechanism of DNA damage response.
Results:Tetrahydrofolic acid exerted cytotoxicity on Adh5-deficient and Fanconi anemia-deficient cells, leading to accumulated DNA damage and chromosome breakage . It induced the formation of intracellular FANCD2 and ser139-H2AX foci in a dose-dependent manner [1]. Tetrahydrofolic acid activated the DNA damage response pathway, depleted nucleotide pools and mediated homologous recombination repair [1].
In vivo
Methods:Adh5-knockout mice received daily intraperitoneal injection of tetrahydrofolic acid at 62.5 mg/kg. Hematopoietic function, protein phosphorylation level and survival of hematopoietic progenitor cells were detected.
Results:Tetrahydrofolic acid treatment disrupted hematopoietic function, increased the phosphorylation level of ser139-H2AX and reduced the survival rate of hematopoietic progenitor cells, indicating that excessive tetrahydrofolic acid possesses mutagenicity and genotoxicity to bone marrow cells [1].
