| Description | STINGantagonist-3 is a potent STING antagonist with an IC50 value of 2.3 nM against human wild-type STING. It effectively inhibits both human wild-type STING and gain-of-function STING mutants N154S and V155M. In stimulated human whole blood, STINGantagonist-3 suppresses the production of IFN-α2a. Additionally, it inhibits IP-10 production in activated human skin microvascular endothelial cells (HMVEC-d). This compound is applicable for research in autoimmune and autoinflammatory diseases, interferonopathies, and fibrotic disorders. |
| In vitro | STING antagonist-3 (Example 21) inhibits human wild-type STING R232 in a competitive HTRF binding assay with an IC50 of 2.3 nM. In a DSF experiment, STING antagonist-3 (100 μM; temperature ramp from 15°C to 95°C over 160 cycles) stabilizes human wild-type STING protein against thermal denaturation, showing a ΔTm of 29 K. Additionally, STING antagonist-3 (10 μM; 60 min pre-incubation; 22 h post-stimulation) suppresses cGAMP-induced IFNα2α production in human whole blood with an IC50 of 0.029 μM. Furthermore, STING antagonist-3 (pre-incubation 1 hour; stimulation 6 hours) inhibits dsDNA-induced IP-10 production in human HMVEC cells with an IC50 of 6.8 nM. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
