STAT3-IN-44 is a potent STAT3 inhibitor with IC50 values of 1.84 μM (C6 cells) and 4.81 μM (A549 cells). It inhibits the phosphorylation of STAT3, downregulates Bcl-2 expression, and upregulates Caspase-3 expression, thereby promoting late-stage apoptosis. STAT3-IN-44 effectively suppresses tumor cell proliferation and migration and is applicable in research on cancers such as glioma and lung cancer.

STAT3-IN-44 (Compound 18q) at a concentration of 10 μM, over 24 hours, significantly induces apoptosis in C6 cells through the STAT3-Bcl-2/Caspase-3 pathway. At concentrations ranging from 2.5 to 10 μM over 10 days, it nearly completely inhibits colony formation of C6 cells at the 10 μM concentration. Additionally, when used at 10 μM for 24 hours, it significantly suppresses C6 cell migration. STAT3-IN-44, applied at 50 μM and temperatures between 37-70℃, markedly enhances the thermal stability of STAT3 in C6 cells.
