| In vitro | SJ1008030 (compound 8) exhibited activity in CRLF2r cell line (MHH-CALL-4) with an IC50 of 5.4 nM after 72-hour treatment with TFA [1]. Dose-dependent degradation of JAK, GSPT1, and IKZF1 was observed in MHH-CALL-4 cells treated with SJ1008030 (0-4.3 μM; 72 hours) TFA [1]. Additionally, SJ1008030 (0-10 μM; 24h) TFA dose-dependently degraded JAK2 and GSPT1 proteins in xenografted bone marrow cells SJBALL021415, indicating inhibition of the JAK-STAT signaling pathway [1]. |
