| In vitro | PROTAC EZH2 Degrader-34, also known as compound E7 (13), is highly effective in inhibiting the enzymatic activity of EZH2, with an IC50 of 6.30 μM. It induces CRBN-mediated proteasomal degradation of PRC2 subunit proteins, reducing H3K27me2/3 levels in WSU-DLCL-2 cells. Furthermore, it demonstrates strong antiproliferative effects in Pfeiffer cells (IC50 of 3.69 μM) and prostate cancer cell lines (IC50 of 0.21 μM). |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
