Description
PROTACCDK2/4/6 Degrader-1 is an orally active CDK2/CDK4/CDK6 PROTAC degrader. It is a prodrug created through a one-step reaction from PROTACCDK2/4/6 Degrader-2 using chloromethylnorvaline ester. This compound is applicable in research related to malignant melanoma.
In vitro
PROTAC CDK2/4/6 Degrader-1 (compound 11) is a prodrug derived from a one-step reaction of PROTAC CDK2/4/6 Degrader-2 (compound 3) with chloromethyl neopentanoate. PROTAC CDK2/4/6 Degrader-2 is a PROTAC CDK2/4/6 degrader that targets the degradation of CDK 2/4/6 proteins and complexes, inducing cell cycle arrest and apoptosis in melanoma cells.
In vivo
PROTAC CDK2/4/6 Degrader (compound 11), administered orally at a dosage of 200 mg/kg once daily for 15 days, significantly inhibits tumor growth and facilitates the degradation of CDK2/4/6 in the B16F10 xenograft mouse model.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
