| In vitro | NLRP3-IN-16 (Compound 12d) inhibits the release of IL-1β with an IC50 of 0.065 μM, as measured by ELISA [1]. At a concentration of 2 μM, for 3 hours, it suppresses the secretion of IL-1β (p17) and caspase-1 (p20) in mouse peritoneal macrophages (PM) [1]. Additionally, NLRP3-IN-16 impedes the formation of the NLRP3 inflammasome complex by inhibiting ASC oligomerization [1]. It exhibits metabolic stability in human and mouse liver microsomes, with a half-life (T 1/2) of 223.5 minutes and an intrinsic clearance (Cl int) of 6.2 μL/min/mg [1]. |

Dissolve 2 mg of the compound in 100 μL DMSO to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
