In vitro
In LPS-primed primary bone marrow-derived macrophages (BMDMs), the chemical NLRP3 inhibitor NLRP3-IN-NBC6 at a concentration of 10 μM and applied for 15 minutes, effectively suppresses the release of IL-1β triggered by Imiquimod. Additionally, NLRP3-IN-NBC6 fully blocks the secretion of NLRP3-dependent IL-1β in neutrophils. These neutrophils, sourced from both wild-type and NLRP3 knockout (KO) mouse bone marrow, were primed with LPS and subsequently treated with Nigericin in the presence of 10 μM NLRP3-IN-NBC6 [1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
