MALT1-IN-13 (compound 10m) is an inhibitor of the mucosa-associated lymphoid tissue lymphoma translocation protein (MALT1), forming a covalent and irreversible bond with the MALT1 protease, thereby inhibiting its activity with an IC50 of 1.7 μM. It suppresses the proliferation of ABC-DLBCL and induces apoptosis in ABC-DLBCL HBL1 cells. Additionally, MALT1-IN-13 regulates the mTOR and PI3K-Akt pathways.

| In vitro | MALT1-IN-13 (0-10 μM) induces apoptosis in HBL1 cells and inhibits the proliferation of ABC-DLBCL HBL1, TMD8, and GCB-DLBCL OCI-LY1 cells, with GI50 values of 1.5, 0.7, and >25 μM, respectively. It downregulates the expression of the MALT1 and NF-kB pathways, while upregulating the mTOR and PI3K-Akt pathways, exerting antitumor effects in HBL1 cells. |
| In vivo | MALT1-IN-13 (25 mg/kg, administered intraperitoneally for 12-14 days) demonstrates tumor-specific activity against MALT1 protease, effectively inhibiting tumor growth in HBL1/TMD8 xenograft NCG mice. |
