LATS-IN-2 is a potent inhibitor of LATS, demonstrating significant inhibitory activity against LATS1 with an IC50 value of 1.3 nM. By inhibiting LATS kinase activity, LATS-IN-2 reduces the phosphorylation of YAP. Additionally, LATS-IN-2 increases the thickness of mouse epidermis and can be used in research on ocular diseases, such as limbal stem cell deficiency and corneal endothelial dysfunction.

| Description | LATS-IN-2 is a potent inhibitor of LATS, demonstrating significant inhibitory activity against LATS1 with an IC50 value of 1.3 nM. |
| In vitro | LATS-IN-2 (Compound 133) significantly reduces the phosphorylation levels of YAP (Ser127) in human HaCaT keratinocytes at a concentration of 9 μM over 1 hour. Additionally, LATS-IN-2 (1-20 μM, 12-16 days) facilitates the expansion of seeded limbal stem cells (LSCs) by 100 to 2200-fold while maintaining p63alpha-positive cells within the expanded population. Furthermore, LATS-IN-2 (0.5-25 μM, 1-2 weeks) induces robust proliferation of human corneal endothelial cells (CECs). |
| In vivo | LATS-IN-2 (Compound 133), applied topically at concentrations of 0.2-2 mg/mL twice daily for 2 days, significantly upregulates the expression of the YAP target gene Cyr61 in a dose-dependent manner in the skin at the wound margins in a full-thickness excision wound model in mice. |
