Bioactivity

Description	KRAS G12D inhibitor 20 (Compound 14) is a selective G12D KRAS inhibitor with antitumor activity.
In vitro	KRAS G12D inhibitor 20 (72 h) exhibits antiproliferative activity against human pancreatic cancer cell lines PANC-1 (KRAS G12D) and BxPC3 (KRASwt), with IC 50 values of 5.5 μM and 8.2 μM, respectively, demonstrating low cytotoxicity to normal cells.
In vivo	KRAS G12D inhibitor 20 (6 mg/kg, intraperitoneal injection, 6 doses) does not significantly affect the body weight of healthy BALB/c female mice. In a xenograft mouse model using immunodeficient SCID male mice carrying KRASG12D (PANC-1), KRAS G12D inhibitor 20 (6 mg/kg, intraperitoneal injection) reduces tumor volume by 13.1%.
