Description
IDN-7314 is a broad-spectrum caspase inhibitor with an IC50 ranging from 0.2 to 7 nM against all tested caspases. It effectively eliminates caspase-3/7 activity induced by Jo2. Additionally, IDN-7314 reduces procoagulant activity of tissue factor in hepatocytes. It is applicable in studies of chemically induced hepatitis, fulminant hepatic failure, and apoptotic liver damage.
In vitro
IDN-7314 is a potent pan-caspase inhibitor, exhibiting IC 50 values ranging from 0.2 to 7 nM across all tested caspases. At a concentration of 100 nM, IDN-7314 completely abolishes Jo2-induced caspase-3/7 activity and significantly reduces the procoagulant tissue factor activity of hepatocytes along with the release of tissue factor-positive microvesicles.
In vivo
IDN-7314 (10 mg/kg; intraperitoneal injection) effectively inhibits hepatocyte apoptosis and caspase-3 activation in female Sprague-Dawley rats induced by galactosamine, with a reduction in apoptotic hepatocytes by 76%-71%, depending on the dosing regimen. However, it only slightly reduces plasma ALT levels and necrotic cell death in this model. Additionally, IDN-7314 (intraperitoneal injection; single dose, 3 mg/kg) inhibits caspase activation, prevents procoagulant changes, and alleviates hepatocellular damage in mice with Fas-induced apoptotic liver injury.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
