Description
HBC-12551 is an orally active BTK inhibitor with an IC50 of 1.31 nM for BTK and 2.18 nM for BTKC481S in HEK293 cells. It exhibits antitumor activity against diffuse large B-cell lymphoma.
In vitro
HBC-12551 effectively inhibits BTK phosphorylation in HEK293 cells with an IC50 of 1.31 nM, as well as the phosphorylation of the BTK C481S mutant with an IC50 of 2.18 nM. In addition, after 72 hours of incubation, HBC-12551 significantly reduces cell viability in malignant B cell lines, including TMD8 (IC50 = 1.40 nM), TMD8-BTK-C481S (IC50 = 1.92 nM), OCI-Ly10 (IC50 = 1.86 nM), and REC-1 (IC50 = 3.11 nM). Moreover, HBC-12551 strongly suppresses B cell activation in human peripheral blood mononuclear cells (PBMCs), with an IC50 of 0.95 nM for CD69 expression following a 30-minute treatment.
In vivo
HBC-12551 (10-20 mg/kg; oral administration; once daily; 14 days) exhibits a potent dose-dependent inhibition of tumor growth in the TMD8 xenograft model.
