Description
Glycolipid C34, an α-GalCer analogue, is a CD1d-binding glycolipid with antimicrobial properties. It activates invariant natural killer T cells and promotes the secretion of IFN-γ and IL-4. In mouse models, Glycolipid C34 aids in the clearance of bacterial infections and enhances protective immunity against viral infections, making it useful in related research on bacterial and viral infections.
In vitro
Glycolipid C34 can activate invariant natural killer T cells through CD1d binding, promoting the secretion of IFN-γ and IL-4.
In vivo
Glycolipid C34, when administered intraperitoneally at a dose of 50 μg/kg as a single injection three hours post-infection, significantly reduces the liver bacterial load in C57BL/6 mice infected with Sphingomonas capsulata. At a dose of 150 μg/kg, administered similarly, it effectively lowers the bacterial load in BALB/c mice with thigh infections of S. aureus Xen29. Immediate treatment within 0-6 hours post-infection significantly decreases the bacterial load, while delayed administration (at 6 hours post-infection) reduces the efficacy. Additionally, Glycolipid C34 (1 μg) given intraperitoneally twice, one day before and one day after infection, enhances survival rates in C57BL/6 mice challenged with JEV and WSN, with its protective effects reliant on intact innate and adaptive immune components.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
