| In vitro | Method: Rat lens homogenate supernatant was used as the crude enzyme source. The reaction system contained 135 mM Na,K-phosphate buffer (pH 7.0), 100 mM Li₂SO₄, 0.03 mM NADPH, and 1 mM DL-glyceraldehyde. Various concentrations of Floramanoside B were added, and the mixture was incubated at 30°C for 30 min. The consumption of NADPH was measured by fluorescence spectroscopy (excitation 360 nm, emission 460 nm) to evaluate the inhibitory activity against aldose reductase (AR). Result: Floramanoside B exhibited inhibitory activity against aldose reductase, with an IC₅₀ value of 13.7 μM [1]. Method: Various concentrations of Floramanoside B were mixed with 100 μM DPPH ethanol solution and 0.1 M acetate buffer (pH 5.5). After 30 min, the change in absorbance was measured at 517 nm, and the DPPH radical scavenging rate was calculated. Result: Floramanoside B exhibited DPPH radical scavenging activity, with an SC₅₀ value of 6.2 μM [1]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
