Description
CXCR6 antagonist1 (Compound 81) is an orally active CXCR6 antagonist. It inhibits the CXCR6 receptor signaling pathways, including β-arrestin recruitment and Forskolin-induced cAMP production. CXCR6 antagonist1 reduces tumor growth in a hepatocellular carcinoma mouse xenograft model and can be utilized in research related to hepatocellular carcinoma.
In vitro
CXCR6 antagonist 1 effectively inhibits CXCL16-induced β-arrestin recruitment in DiscoveRx cells expressing CXCR6, with an IC50 of 40 nM. It also suppresses forskolin-induced cAMP signaling in the same cell type, with an IC50 of 540 nM. Additionally, CXCR6 antagonist 1 dose-dependently inhibits the migration of hepatocellular carcinoma SK-HEP-1 cells over 72 hours, with an IC50 of 2.1 μM.
In vivo
Administering CXCR6 antagonist 1 at a dosage of 30-60 mg/kg through oral gavage daily from day 4 to day 30 results in a significant reduction of tumor weight in female NOD/SCID mice with SK-HEP-1 liver cancer.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
