  • Modeling Mechanism:

    Bovine Type II Collagen, as a foreign antigen, induced mice to produce anti-type II collagen-specific IgG2a antibodies after emulsification with complete Freund's adjuvant (FCA), activating an autoimmune response. By activating the NF-κB and MAPK (ERK1/2, p38) signaling pathways, it promoted the release of pro-inflammatory factors TNF-α and IL-6, leading to neutrophil infiltration, cell proliferation, and joint damage in joint tissues.

    Mice, DBA/1, Male, 8 weeks old

    Dosage and Administration Route:

    ① Primary immunisation: Bovine Type II Collagen, emulsified in complete Freund's adjuvant (FCA), subcutaneous injection (Day 0);
    ② Booster immunisation: Same dose Bovine Type II Collagen+FCA, Subcutaneous injection (Day 21);
    ③ Intervention control (optional): 10 mg/kg high-density lipoprotein (HDL), Intravenous injection via tail vein (from Day 28)

    Dosing Frequency and Duration Model:

    Primary and booster immunisations: single dose; Intervention control: once every 3 days for 5 doses total

  • Validation:

    Clinical phenotype: Significant joint swelling (increased claw thickness) appeared after day 28, with elevated arthritis scores (up to 10 points) and weight loss; Molecular markers: Serum TNF-α and IL-6 levels were significantly elevated (TNF-α: 112.6±9.373 pg/mL, IL-6: 72.74±8.803 pg/mL), and anti-type II collagen IgG2a antibody levels were upregulated; Histological markers: HE staining of joint tissue showed neutrophil infiltration, cell proliferation, disordered arrangement, and joint structural destruction; Signaling pathways: Significantly elevated expression of NF-κB p65, p-IκBα, and MAPK pathway p-ERK1/2 and p-p38 proteins in joint tissue.

*Precautions: They were put to death 60 days after the model was created.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
