| In vitro | For the BODIPY-cholesterol assay:Reconstitute the analogue in DMSO to prepare a stock solution of BODIPY-cholesterol at a concentration of 1-10 μM[2]. Dilute cells with PBS (or other buffers), wash them (300×g, 10 min) to eliminate excess diluent, and resuspend the loose pellet with vehicle media to achieve the desired concentration[2].Label cell suspensions with BODIPY-cholesterol (final concentration of 0.5-5 μM), ensuring thorough mixing, and incubate for 10-20 minutes at 37 °C[1].Dilute the cells with each of the capacitation media conditions, as outlined in Incubation media, to a final concentration. Incubate for 2 hours before flow cytometric assessment[2]. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
