  • Modeling Mechanism:

    Angiotensin II human TFA enhances vascular receptor affinity by binding to specific receptors on the surface of vascular smooth muscle cells, promoting vasoconstriction and increasing peripheral resistance. At the same time, it activates the sympathetic nervous system and regulates sodium balance (high sodium intake can further enhance receptor sensitivity), inducing persistent hypertension and mimicking the pathological process of clinical renin-angiotensin system activation related to hypertension.

  • Modeling Method:

    Experimental Subject:

    Rats, Sprague-Dawley (SD), Male, Body weight 150–250 g

    Dosage and Administration Route:

    ① Core modelling: Angiotensin II human TFA, Dose adjusted as required for experiment (for receptor blockade studies, exogenously administered at 50 ng per dose to validate blockade effect), Dissolved in sterile physiological saline, Intravenous injection (i.v.);
    ② Dietary intervention:
    - High-sodium group: Standard diet+0.9% saline drinking water;
    - Normal sodium group: Standard diet (containing 0.42% sodium)+conventional drinking water;
    - Low-sodium group: Sodium-free diet+conventional drinking water;
    ③ Control treatment: Control group administered equal volume saline solution, with all other conditions identical;
    ④ Combined Preparation of Hypertensive Models:
    - Bilateral renal artery clamping (Goldblatt): Silver clamp applied to left renal artery, contralateral kidney preserved;
    - Unilateral nephrectomy (Goldblatt): Left renal artery ligated with silver clip, contralateral kidney removed;
    - DOCA hypertension: Subcutaneous implantation of 25 mg DOCA pellets (every 4 weeks)+high-sodium diet;

    Dosing Frequency and Duration Model:

    Single administration; dietary intervention commenced after 6 weeks

  • Validation:

    1. Blood Pressure Indicators: - Baseline Blood Pressure: Normal rat systolic blood pressure is 120-130 mmHg; after modeling, it reaches 175-195 mmHg in the two-kidney-one-clip type and over 210 mmHg in the one-kidney-one-clip type; spontaneously hypertensive rats reach 195-200 mmHg. - Receptor Blocking Verification: After injection of Ang II antibody, blood pressure can decrease by 20-50 mmHg (significant in the two-kidney-one-clip type, no significant decrease in the one-kidney-one-clip type). 2. Receptor Affinity: The high-sodium intake group requires 8 times more Ang II antibody than the low-sodium group to block the vascular response, proving that receptor sensitivity is positively correlated with sodium intake. 3. Metabolic Indicators: Plasma Ang II levels (approximately 150 pg/ml in the low-sodium group, < 20 pg/ml in the high-sodium group) show an inverse relationship with receptor affinity. 4. Specificity Verification: The antibody only blocks the pressor effect of Ang II and does not affect the blood pressure response to norepinephrine.
