ALOX5 stabilizer-1 is an inhibitor of arachidonic acid 5-lipoxygenase (ALOX5) with a Kd value of 7.26 μM. It reduces protein levels of β-catenin and subsequently inhibits the STAT3 signaling pathway. ALOX5 stabilizer-1 induces apoptosis in lung cancer cells and hampers their migration and proliferation, making it useful for research on non-small cell lung cancer (NSCLC).

| In vitro | ALOX5 stabilizer-1 (Compound 10d) effectively inhibits the proliferation of NCI-H820 cells with an IC 50 of 0.11 μM over 72 hours, while exhibiting lower toxicity on 3T3 cells with an IC 50 of 0.48 μM. In a dose-dependent manner, ALOX5 stabilizer-1 (0-0.2 μM, 10 days) suppresses colony formation of NCI-H820 cells. Additionally, at concentrations of 0-0.2 μM over 48 hours, it inhibits NCI-H820 cell migration and induces apoptosis. The compound directly targets the ALOX5 protein, enhancing its stability and consequently decreasing β-catenin protein levels, ultimately inhibiting the STAT3 signaling pathway in NCI-H820 cells. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
