Description
TGF-β/Smad-IN-2 is an orally active TGF-β/Smad inhibitor. It suppresses TGF-β-induced differentiation of fibroblasts into myofibroblasts and collagen deposition, with an IC50 value of 102 μM for reducing collagen in LL29 cells. TGF-β/Smad-IN-2 acts by modulating the SMAD3/SMAD7 signaling pathway and decreases lung index in mice with pulmonary fibrosis. This compound is applicable in pulmonary fibrosis research.
In vitro
TGF-β/Smad-IN-2 (Compound 10b) at concentrations of 50-100 μM over 48 hours significantly reduces the expression of fibrosis markers (FN1, α-SMA, COLLAGEN1α1, COLLAGEN3α1, TIMP1, VIMENTIN) at the mRNA level, and it inhibits the differentiation of fibroblasts into myofibroblasts induced by TGF-β in LL29 and DHLF cells. Additionally, TGF-β/Smad-IN-2 decreases the protein levels of α-SMA, COLLAGEN3α1, and p-COFILIN in DHLF cells while restoring E-CADHERIN expression in response to TGF-β stimulation.
In vivo
TGF-β/Smad-IN-2 (Compound 10b) administered at 150-300 mg/kg via gavage once daily for 14 days, enhances weight recovery and survival rates, and reduces the lung index in C57BL/6 mice with Bleomycin (BLMN)-induced pulmonary fibrosis.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
