Description
TAT-GluN2BCTM is a membrane-permeable peptide that selectively targets active DAPK1 (Death-associated protein kinase 1) for lysosomal degradation, protecting neurons against oxidative stress and NMDAR-mediated excitotoxicity, making it valuable for neuroprotection research studies [1].
In vitro
TAT-GluN2BCTM (200 µM) reduces DAPK1 levels in NMDA-treated primary neuronal cultures in a time-dependent manner over 0.5, 2, and 4 hours [1].
In vivo
Administration of TAT-GluN2BCTM (10 mg/kg; i.v.; single) resulted in a significant decrease in the levels of active DAPK1 in the ischemic hemisphere of the rat brain [1]. TAT-GluN2BCTM demonstrated substantial neuroprotective effects in the cortex and striatum of rats [1]. Furthermore, TAT-GluN2BCTM was able to specifically reduce the activity of the active (but not inactive) form of DAPK1 [1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
