| In vitro | Smurf-1-IN-3 (compound 45) effectively inhibits the self-ubiquitination of recombinant Smurf-1 with an IC50 of 0.0022 μM when tested at a concentration of 0.0022 μM for 45 minutes. Furthermore, Smurf-1-IN-3 (compound 45), within a range of 6.75 nM to 22.5 μM overnight, stabilizes the Smurf-1 protein in tetracycline-inducible HEK293 cells, confirming its ability to inhibit Smurf-1 activity at the cellular level. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
