Perforin-IN-3 is a specific perforin inhibitor that prevents perforin-mediated pore formation in cell membranes and subsequent cell lysis. It effectively inhibits the cytotoxic effects of NK cells on target cells such as leukemia without interfering with FasL- or TRAIL-mediated cell death pathways. Perforin-IN-3 does not affect pneumolysin and can be utilized for studying autoimmune diseases, allograft rejection, graft-versus-host disease, and familial hemophagocytic lymphohistiocytosis [FHL].

Description
Perforin-IN-3 is a specific perforin inhibitor that prevents perforin-mediated pore formation in cell membranes and subsequent cell lysis. It effectively inhibits the cytotoxic effects of NK cells on target cells such as leukemia without interfering with FasL- or TRAIL-mediated cell death pathways. Perforin-IN-3 does not affect pneumolysin and can be utilized for studying autoimmune diseases, allograft rejection, graft-versus-host disease, and familial hemophagocytic lymphohistiocytosis [FHL].
In vitro
Perforin-IN-3 (compound 1) inhibits perforin-mediated lysis of Jurkat T lymphoma cells with an IC 50 (4 h) of 2.7 μM. It reduces the KHYG-1 NK cell-mediated killing rate of K562 leukemia target cells by 50%, and after a 24-hour exposure, the survival rate of KHYG-1 NK cells remains at 72%. At a concentration of 20 μM for 30 minutes, Perforin-IN-3 does not inhibit Fas ligand or TRAIL-mediated death of Jurkat T lymphoma cells. Additionally, it suppresses primary human NK cell-mediated lysis of K562 leukemia target cells by more than 70%.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 µL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 µL Tween 80 and mix well until fully clarified.

3) Add 450 µL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
