| In vitro | PDIA1-IN-1 (Compound P11) inhibits the formation of ASC-GFP specks in lipopolysaccharide-pretreated THP1 monocytes with an IC50 of 890 nM. In a dose-dependent manner, PDIA1-IN-1 (1.25-2.5 μM, 2 h) binds to PDIA1 in these cells. It reduces IL-1β secretion in LPS-pretreated THP1 monocytes at concentrations of 5-10 μM for 1-16 hours. Additionally, 1 μM of PDIA1-IN-1 for 2 hours can restore cell viability, whereas 10 μM for 1 hour decreases cell viability in LPS-pretreated THP1 monocytes. |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
