Description
Macimorelin acetate is an orally active agonist of the GHSR (Growth Hormone Secretagogue Receptor) and functions as a growth hormone (GH) secretagogue. As a ghrelin mimetic, it potently stimulates the release of endogenous growth hormone by binding to GHSR. Macimorelin acetate is clinically utilized as a diagnostic tool for Adult Growth Hormone Deficiency (AGHD) and is being investigated for the treatment of Cancer Anorexia-Cachexia Syndrome (CACS). Additionally, it has shown potential in neurological research for suppressing seizure activity.
In vitro
NFATc1-IN-1 (0、0.5、1.0、1.5、2.0、2.5 μM, 4 days) exhibits potent inhibitory activities on osteoclast formation and function, which eventually translates into decreased bone resorption while showing no cytotoxic effects towards osteoclast precursor cells at concentrations of as high as 2.5 μM.[1]
NFATc1-IN-1 (1.5-2.5 μM, 24 h) blocks NFATc1 nuclear translocation and decreases the level of NFATc1.[1]
In vivo
Macimorelin acetate suppresses acute seizures in epileptic mice (5 mg/kg, i.p.) but lacks long-term antiepileptic effects [2]. Orally, it rapidly and dose-dependently increases GH levels, serving as an effective diagnostic for AGHD [1].
